    You can buy it at the DLR ticket booths anytime or day before the day you want to use it.

    There is nowhere in Downtown Disney that sells one day tickets, the World of Disney store does sell multi-day tickets and Annual Pass tickets.

    Your hotel MIGHT sell One day tickets and/or multi-day tickets... most Good Neighbor Hotels have multi-day tickets, places like the new Doubletree Suites and the Hilton have what are called "Disney Desks", which are run by Walt Disney Travel, these special locations (there is also one in South Coast Plaza) have wide variety of tickets, and your best bet for advance purchase one day tickets.
